Inside The One: A Closer Look At Our Longevity Ingredients

Longevity formulations tend to be judged by their headline ingredient, but the more interesting story is usually in the supporting cast. Here’s a review of what the research says about each ingredient in our blend.

Nicotinamide mononucleotide (NMN). Directly replenishes NAD+, the coenzyme essential for mitochondrial energy production, DNA repair, and cellular metabolism. NMN is a direct precursor to NAD+, a coenzyme every cell needs for energy production and DNA repair, and one that reliably declines with age. Across more than a dozen human trials, oral NMN has consistently and measurably raised blood NAD+ levels. 

Creatine monohydrate. Creatine regenerates ATP in neurons and muscle cells, demonstrably enhancing working memory, reducing cognitive fatigue, and maintaining brain energy reserves as you age. Creatine is the most thoroughly researched compound in sports nutrition, with decades of consistent evidence for increasing strength, power output and lean mass, particularly alongside resistance training. What's newer is research into ageing and cognition: creatine appears to help preserve muscle mass in older adults, and a growing body of work on the "muscle-brain axis" suggests cognitive benefits, especially under conditions like sleep deprivation. 

L-theanine. Generates relaxed alertness without drowsiness. Lowers stress hormones, increases alpha wave activity, and produces sustained mental performance throughout demanding days. Found naturally in tea, L-theanine has good trial evidence for promoting a calm, alert mental state and blunting the body's physiological stress response, including measurable reductions in heart rate and cortisol during acute stress. Trials in middle-aged and older adults also report improvements in working memory and attention. 

Resveratrol. Extracted from Japanese Knotweed. Triggers sirtuin proteins that regulate cellular survival, repair, and lifespan. The polyphenol that positioned red wine in longevity discourse - now delivered at research-validated concentrations.

Glycine. This amino acid is a building block for collagen and acts as a calming neurotransmitter in the nervous system. Its best-supported use is sleep: modest doses have been shown to improve sleep quality and reduce next-day fatigue in people with disrupted sleep.

Sodium hyaluronate. Oral hyaluronic acid has solid trial support: multiple randomised, placebo-controlled studies show measurable improvements in skin hydration and barrier function over 8 to 12 weeks, with some evidence of improved elasticity.

Magnesium (as glycinate). Magnesium is a cofactor in over 300 enzymatic reactions, spanning energy production, nerve transmission and muscle function. The glycinate form is generally well absorbed. Evidence here mostly comes from decades of nutritional science establishing magnesium's essential roles.

Vitamin C (ascorbic acid). Vitamin C's roles in collagen synthesis, iron absorption and antioxidant defence are well established.

Vitamin D3 (colecalciferol). Vitamin D governs calcium absorption and bone mineralisation, and has a well-documented regulatory role in immune function. Deficiency is common in adults with limited sun exposure, making adequate supplementation very useful for a meaningful portion of the population.

Vitamin K2 (as MK-7). K2 works alongside vitamin D to direct calcium into bone rather than soft tissue, supporting bone density over time.

Thiamine (vitamin B1). An essential cofactor in carbohydrate metabolism and nerve function, thiamine's inclusion here is primarily about ensuring sufficiency.

Pyridoxal-5-phosphate (active vitamin B6). This is the biologically active form of B6, involved in amino acid metabolism and the synthesis of neurotransmitters including serotonin and dopamine.

Mecobalamin (active vitamin B12). B12 is essential for red blood cell formation and nervous system function. Mecobalamin is one of the coenzyme forms the body uses directly, which may be a genuine advantage for people with absorption difficulties or higher needs, such as older adults.

Quatrefolic (active folate, as 5-MTHF). This active folate form raises blood folate and lowers homocysteine reducing fatigue and supporting energy levels.

Choline (as bitartrate). Choline is a precursor to acetylcholine, a neurotransmitter involved in memory and muscle control, and plays a role in liver lipid metabolism.

Biotin. Biotin supports energy metabolism as a cofactor for several key enzymes.

Zinc (as citrate). Zinc is essential to immune cell function and acts as a cofactor for antioxidant enzymes in the body. It has a solid evidence base, particularly for immune support, and deficiency is common.
